Telomeres and Telomerase in Cancer: Overview and Therapeutic Potential

نویسندگان

چکیده

Telomeres are specialized structures of eukaryotic chromosomes that protect the tips chromosomes. prevent from losing base pair sequences at their ends, and also stop fusing together. Telomerase is an enzyme rebuilds telomeres maintains genomic stability by extending after each cell division. activity prevents cellular aging, known as senescence. Immortal lines can be created inducing expression telomerase in cells normally lack telomerase. Cells need to maintain integrity mutations lead cancer. Such activation mentioned above often cancer development preserving rapidly dividing cells. If a keeps overcomes limitations telomeres, cancerous tumor form. In healthy cells, achieve balance between limiting lifespan keeping growth bay.
It has been established biomarker tumorigenesis. This makes attractive target for therapy, studies indicate methods treatment such oligonucleotides, immunotherapies, stabilization G-quadruplexes all hold immense possibilities field oncology. discoveries provide insight on potential play significant role prevention before tumorigenesis, or diagnosis. Therefore, viable therapeutic target, area interest study.

Telomeres and telomerase in cancer.

Myriad genetic and epigenetic alterations are required to drive normal cells toward malignant transformation. These somatic events commandeer many signaling pathways that cooperate to endow aspiring cancer cells with a full range of biological capabilities needed to grow, disseminate and ultimately kill its host. Cancer genomes are highly rearranged and are characterized by complex translocatio...

Role of telomeres and telomerase in cancer.

There is mounting evidence for the existence of an important relationship between telomeres and telomerase and cellular aging and cancer. Normal human cells progressively lose telomeres with each cell division until a few short telomeres become uncapped leading to a growth arrest known as replicative aging. In the absence of genomic alterations these cells do not die but remain quiescent produc...

Treating Cancer by Targeting Telomeres and Telomerase

Telomerase is expressed in more than 85% of cancer cells. Tumor cells with metastatic potential may have a high telomerase activity, allowing cells to escape from the inhibition of cell proliferation due to shortened telomeres. Human telomerase primarily consists of two main components: hTERT, a catalytic subunit, and hTR, an RNA template whose sequence is complimentary to the telomeric 5'-dTTA...

Telomeres and telomerase.

Telomeres are the protective DNA-protein complexes found at the ends of eukaryotic chromosomes. Telomeric DNA consists of tandem repeats of a simple, often G-rich, sequence specified by the action of telomerase, and complete replication of telomeric DNA requires telomerase. Telomerase is a specialized cellular ribonucleoprotein reverse transcriptase. By copying a short template sequence within ...
